Advances on Inverse Metabolic Engineering

  • 摘要: 反向代谢工程通过构建具有遗传多样性的突变文库,结合有效的高通量筛选技术,从而获得由多基因控制的复杂表型。目前,它已成为工业生物技术中研究生物代谢途径和改造菌株性能的强有力工具。反向代谢工程研究中的一个瓶颈问题是如何获得多样化的细胞表型突变库,该文介绍了几种经典的和近期发展起来的解决该瓶颈问题的方法,并列举了这些方法的最新应用进展。

     

    Abstract: Inverse metabolic engineering is a kind of new genetic engineering, which can get complex phenotypes regulated by multiple genes by constructing a library of cells with genetic diversity, following with efficient high throughput screen. Currently, it has become a powerful tool for researching biological metabolic pathways and modifying the biological properties of strains in industrial biotechnology. One of the bottleneck problems in inverse metabolic engineering is how to obtain a variety of cell phenotype mutant library. This paper briefly introduced several classic and recently developed ways to solve the bottleneck problem, and listed the latest research progresses of these methods.
